AIRE-1 Antibody (C-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects AIRE-1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). AIRE-1 (C-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. AIRE-1 protein, also known as autoimmune regulator type 1, plays a crucial role in the immune system by regulating self-antigen expression in the thymus, which is essential for self-tolerance development and autoimmune disease prevention. AIRE-1 is localized in medullary epithelial cells and monocyte-dendritic cells of the thymus, where AIRE-1 orchestrates transcriptional control of genes necessary for proper thymic environment establishment. This localization ensures developing T cells are exposed to diverse self-antigens, allowing autoreactive T cell elimination during negative selection. Mutations in AIRE-1 can lead to impaired thymic T cell selection, contributing to autoimmune conditions like autoimmune polyendocrinopathy candidiasis ectodermal dystrophy (APECED). Using anti-AIRE-1 antibody (C-2) in research can provide insights into immune tolerance mechanisms and AIRE-1 dysfunction implications in autoimmune diseases.

Hello, What do you use as a negative control for AIRE-1, clone C-2? Would you be able to provide an aliquot for our lab? Thanks, Paige

Thank you for your question. We recommend using a normal mouse IgG as a negative control when using this antibody. We do not have a specific negative control cell line. Unfortunately we do not offer a sample of the normal mouse IgG, but we would be happy to send you a sample of the antibody, sc-373703. I am using AIRE-1 (C-2): sc-373703 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Rated 5 out of 5 by from Good antibodyThe antibody works very well in IHC on FFPE. The best dilution is 1:1500 using as antigen retrival EDTA buffer pH 8.0 in water bath at 98°C. The staining is strong and nuclear.
