BAF Antibody (A-11) is a mouse monoclonal IgG2a kappa light chain antibody that detects BAF protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-BAF antibody (A-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Barrier-to-autointegration factor (BAF) plays a critical role in the nucleus, where BAF is primarily located, as BAF binds non-specifically to double-stranded DNA and is essential for maintaining chromatin structure. This localization is vital because BAF facilitates the recruitment of nuclear proteins through interaction with the LEM domain, which is crucial for chromatin decondensation and membrane recruitment during nuclear assembly. Furthermore, BAF′s ability to compress chromatin structure is significant for regulating gene expression in a tissue- or cell type-specific manner, as BAF interacts with various homeodomain transcription factors. Notably, retroviruses such as HIV-1 exploit BAF from host cells, incorporating BAF into preintegration complexes to prevent autointegration of retroviral DNA, thereby enhancing the integration of viral DNA into the host chromosome.

When should the TransCruz reagent sc-166324 X be used instead of the standard form sc-166324?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.

Rated 1 out of 5 by from not suitable for immunofluorescenceAntibody was tested at 1:100 on formaldehyde-fixed HeLa cells and only produced cytoplasmic background. GFP-BAF was transfected as reference, staining for the ER protein REEP5 served as positive control for antibody staining.

Rated 5 out of 5 by from Good BAFI use this antibody at a dilution of 1:3000 in TBS-Tween for 1hr and is enough for an good western blot signal. Loading of 10 ul of total samples is recommended for quantification.
