PARP-1 Antibody (5A5) is a mouse monoclonal IgG1 antibody raised against a fragment of human PARP-1 and is reactive with mouse, rat, and human samples, making anti-PARP-1 antibody (5A5) suitable for detection by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). PARP-1, also referred to as poly(ADP-ribose) polymerase-1, ARTD1, Adprt, and Parp1, is a nuclear DNA-binding protein that plays a critical role in DNA repair, chromatin remodeling, and apoptosis. As a nuclear protein, PARP-1 is primarily located within the nucleus, where PARP-1 is crucial for maintaining genomic stability by recognizing DNA damage and facilitating repair processes. PARP-1′s nuclear localization allows direct binding to DNA strand breaks, where PARP-1 catalyzes the transfer of ADP-ribose from NAD+ to various nuclear proteins, including histones and other chromatin-associated proteins, to modulate chromatin structure and signal the need for repair. This activity is particularly important in response to genotoxic stress, as PARP-1 helps prevent mutations and other deleterious effects of unrepaired DNA damage. Furthermore, cleavage of PARP-1 by caspases during apoptosis generates a p89 fragment that translocates to the cytoplasm, marking a key step in programmed cell death. Deficiencies in PARP-1 are linked to increased chromosomal instability, highlighting PARP-1′s essential function in preserving genome integrity. PARP-1 monoclonal antibody (5A5) enables researchers to effectively study the intricate mechanisms of DNA repair, the maintenance of genomic integrity, and apoptosis, making this tool vital for advancements in molecular biology and medical research.
