  • specific for an epitope mapping between amino acids 1-27 at the N-terminus of PARP of mouse origin
  • recommended for detection of full-length PARP-1 and the N-terminal cleavage product of PARP-1 of mouse, rat and human origin by WB, IP, IF and ELISA
  • TransCruz reagent for ChIP application (sc-365315 X, 200 µg/0.1 ml)
  • See PARP1 (F-2): sc-8007 for PARP1 antibody conjugates, including AC, HRP, FITC, PE, Alexa Fluor® 488, 594, 647, 680 and 790.
  • m-IgG Fc BP-HRP and m-IgG1 BP-HRP are the preferred secondary detection reagents for PARP1 Antibody (D-1) for WB applications. These reagents are now offered in bundles with PARP1 Antibody (D-1) (see ordering information below).

SEE ALSO...

PARP-1 Antibody (D-1) is a mouse monoclonal IgG1 kappa light chain antibody that targets an epitope between amino acids 1-27 at the N-terminus of PARP-1, with reactivity in mouse, rat, and human samples, making it suitable for applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PARP-1, also called poly(ADP-ribose) polymerase-1, ARTD1, Adprt, and Parp1, is a key nuclear protein that regulates DNA repair, chromatin remodeling, and apoptosis. PARP-1 interacts with several proteins essential to its function. One significant interaction is with DNA-dependent protein kinase (DNA-PK), which works alongside PARP-1 in facilitating non-homologous end joining, a crucial pathway for repairing double-strand DNA breaks. PARP-1 also interacts with histones, modifying them through poly(ADP-ribosyl)ation to relax chromatin and allow access to damaged DNA. Additionally, PARP-1 complexes with XRCC1, a scaffolding protein involved in single-strand break repair, stabilizing repair machinery at sites of damage. PARP-1′s interaction with apoptosis-inducing factor (AIF) is another vital connection, as AIF translocates from mitochondria to nucleus in a PARP-1-dependent manner to facilitate chromatin condensation and large-scale DNA fragmentation during programmed cell death. These interactions highlight PARP-1′s central role in maintaining genomic stability, ensuring proper DNA repair, and regulating cell death, particularly in response to genotoxic stress. Ultimately, anti-PARP-1 antibody (D-1) is a highly reliable and versatile tool that empowers researchers to explore PARP-1′s essential interactions and functions in DNA repair, chromatin remodeling, and apoptosis, advancing genomic stability understanding across multiple species and experimental platforms.

What application is the blocking peptide sc-365315 P appropriate for?

Asked by: Cweed
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization
