PUS1 Antibody (C-4) is a mouse monoclonal IgG2b kappa light chain antibody that detects PUS1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-PUS1 antibody (C-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PUS1, or pseudouridine synthase 1, is a crucial enzyme that catalyzes the conversion of uridine to pseudouridine in RNA, a modification that enhances the stability and functionality of tRNA and rRNA molecules. This post-translational modification is vital for proper protein synthesis and cellular function, as pseudouridine improves the structural integrity of RNA, thereby influencing the efficiency of translation and overall fidelity of gene expression. PUS1 is predominantly located in the nucleus, where PUS1 forms complexes with other proteins, such as retinoic acid receptor gamma (RARG) and SRA1 RNA, underscoring PUS1′s role in RNA processing and regulation. PUS1 expression is widespread, with particularly high levels found in brain and skeletal muscle tissues, and mutations in the PUS1 gene are associated with myopathy with lactic acidosis and sideroblastic anemia (MLASA), a rare genetic disorder affecting oxidative phosphorylation in muscle and bone marrow. The deduced human PUS1 protein consists of 348 amino acids and exhibits 92% sequence homology with its mouse counterpart, highlighting the evolutionary conservation of PUS1′s function across species.

Can PUS1 (C-4): sc-376442 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

    Asked by: Dr Ninau Qelp
Thank you for your question. Yes, PUS1 (C-4): sc-376442 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
