Uev1A/Mms2 Antibody (E-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects Uev1A/Mms2 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Uev1A/Mms2 antibody (E-6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Uev1A, also known as ubiquitin-conjugating enzyme E2 variant 1 (UEV1) or CROC1, and Mms2 (UEV2) share sequence similarities and predicted structural features with ubiquitin-conjugating enzymes, or E2s, but lack a critical cysteine residue necessary for catalytic activity typical of E2 enzymes. While Uev1A shows no ubiquitin-conjugating activity in vitro, exogenous Uev1A expression in colon carcinoma cells inhibits differentiation upon confluence. Nuclear localization of Uev1A, excluding nucleolar regions, enables collaboration with TRAF6, a RING domain protein, to facilitate synthesis of unique polyubiquitin chains linked through Lysine 63 of ubiquitin. Uev1A interaction with UBC13 (ubiquitin-conjugating enzyme E2N) suggests involvement in protein degradation, particularly in muscle and testis tissues. In yeast, Mms2/UBC13 complex assembles novel polyubiquitin chains critical for DNA repair signaling, suggesting UEV proteins enhance ubiquitin conjugation diversity and selectivity in cellular signaling and stress responses.
