Estrogen Receptor alpha Antibody (10H12B10) is a mouse monoclonal IgG1 kappa light chain engineered to detect human Estrogen Receptor alpha (ERα) in western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA) assays. Anti-ERα antibody (10H12B10) is raised against a recombinant protein corresponding to amino acids 301-595 of human ERα, targeting a critical region within the ligand-binding domain that is essential for the receptor′s function as a ligand-activated transcription factor involved in regulating the normal activities of reproductive tissues. ERα (10H12B10) monoclonal antibody recognizes a protein subject to various post-translational modifications, including phosphorylation, acetylation, and ubiquitination, which play pivotal roles in modulating activity, stability, and interactions with co-regulatory proteins and DNA. Phosphorylation of ERα can enhance or inhibit transcriptional activity and alter responsiveness to estrogen, while ubiquitination typically governs receptor degradation and turnover, thereby maintaining cellular homeostasis. ERα monoclonal antibody (10H12B10) supports comprehensive investigations into the distinct regulatory mechanisms of ERα and ERβ, their differential activation by various ligands, and their crucial roles in cellular signaling pathways and gene expression regulation.

ER alpha (10H12B10) antibodies is it available for use in immunohistochemistry or can it be produced

Thank you for your question. We do not currently recommend this specific clone for immunohistochemistry (IF or IHC(P)). There are several different monoclonal antibodies against ERalpha that we have tested and do recommend for this application. Please contact our Technical Service department for a product recommendation.

How much sample do you recommend loading on the gel for Western blot detection using ERα (10H12B10): sc-130072?
