ERK 2 Antibody (D-2) is a mouse monoclonal IgG2b kappa light chain antibody raised against amino acids 1-286 of ERK2 of human origin, specifically recognizing the p42 isoform of ERK2 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin embedded sections (IHC/P), flow cytometry (FCM), and enzyme-linked immunosorbent assay (ELISA). ERK2, also known as Mitogen-Activated Protein Kinase 1 (MAPK1), is a pivotal component of the MAPK/ERK signaling pathway, which transmits extracellular signals from growth factors, hormones, and neurotransmitters to elicit cellular responses. Activation of ERK2 occurs via phosphorylation by upstream kinases like MEK1/2, leading to dimerization and translocation into the nucleus. Once in the nucleus, ERK2 phosphorylates various transcription factors and regulatory proteins, influencing gene expression related to cell proliferation, differentiation, survival, and development. Dysregulation of ERK2 activity has been implicated in numerous pathological conditions, including cancer and neurodegenerative diseases, making ERK2 a significant focus in biomedical research. ERK 2 (D-2) antibody′s broad reactivity across species makes this product an invaluable tool for researchers investigating the critical roles of ERK2 in cell signaling and potential as a therapeutic target and research agent.

Hi, does this ERK2 ab detect both unphosphorylated and MEK-phosphorylated ERK2?

Thank you for your question. Yes, this is a total ERK 2 antibody and can detect both phosphorylated and un-phosphorylated ERK 2. Please refer to the Western blot data on our website for further evidence of this reactivity.

What is the evidence that this mAb is actually specific to ERK2 and does *not* bind (or minimally binds) ERK1? The given immunizing sequence (ERK2 345-358) shares 12/13 amino acid residues with ERK1 so it is a little hard to believe this is ERK2-specific.

Thank you for your inquiry. Since we do not epitope map our monoclonal antibodies, we cannot say with certainty if this clone is cross-reactive with ERK 1, but there is a good possibility that it is reactive with ERK 1 p44.
