PDPK1 Antibody (E-3) is a mouse monoclonal IgG2a kappa light chain antibody that detects PDPK1 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PDPK1 (E-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates. PDPK1, also known as 3-phosphoinositide dependent protein kinase 1, is a 556 amino acid protein that is ubiquitously expressed and primarily localized to the cell membrane, cytoplasm, and nucleus. As a master kinase, PDPK1 plays a crucial role in phosphorylating and activating a specific subgroup of the AGC family of protein kinases, which are essential for various cellular processes. PDPK1 is vital for mediating signal transduction pathways that control the proliferation, survival, and growth of developing pancreatic beta cells, as well as regulating calcium uptake and calcium-activated potassium channels in mast cells. Additionally, PDPK1 is involved in the regulation of chemotaxis and motility of vascular endothelial cells, cardiac homeostasis, and thymocyte development. Belonging to the protein kinase superfamily, PDPK1 features a PH domain that is critical for homodimerization, localization, and nuclear import, alongside a protein kinase domain. The gene encoding PDPK1 is located on human chromosome 16p13.3 and gives rise to five alternatively spliced isoforms, highlighting the complexity and regulatory potential of this important protein in cellular signaling.

Can PDPK1 (E-3): sc-17765 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, PDPK1 (E-3): sc-17765 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 5 out of 5 by from Outstandingly good antibody for Immunoblot!The SCBT PDPK1 (3-Phosphoinositide Dependent Protein Kinase 1) antibody ((E-3): sc-17765) is excellent for immunoblotting. Multiple isoforms for this protein is expressed (see Uniport entry for this protein). This antibody recognized two isoforms/PTM forms at ~60-50kDa in HEK293 and HepG2 cells lysates which are differentially expressed (see attached blot). Multiple isoforms for this gene has been reported previously, see Stephens et al. 1998 (PMID: 9445477). Stephens et al reported expression of ~60kDa PDPK1 in Cos7 cells. Excellent antibody, highly recommend it for your research.
