Syk Antibody (0.T.142) is a mouse monoclonal IgG2a antibody that detects Syk in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). Syk, or spleen tyrosine kinase, is a 635 amino acid protein characterized by one protein kinase domain and two SH2 domains, and plays a crucial role in B cell signaling. Specifically, Syk acts as a positive effector of B cell antigen receptor (CD79)-stimulated responses, facilitating calcium ion movement through two distinct phospho-regulated pathways. When unphosphorylated, Syk enables calcium ions to traverse a phosphoinositide 3-kinase-dependent pathway, while phosphorylation at Tyr 348 and Tyr 352 shifts calcium flow to a phospholipase C γ-dependent pathway. This regulation of calcium movement is vital for various cellular processes, including differentiation, phagocytosis, proliferation, and B cell development. Notably, Syk expression is upregulated in T cell lymphoma, indicating potential involvement in tumorigenesis. Additionally, Syk exists in two isoforms, short and long, resulting from alternative splicing events, which may have distinct functional implications in cellular signaling pathways.
