SEE ALSO...

IFN-α Antibody (F-7) is a mouse monoclonal IgG2b kappa light chain antibody that detects the IFN-alpha protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). IFN-α (F-7) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. IFN-alpha protein plays a crucial role in immune response, particularly in defense against viral infections, by activating immune cells and enhancing expression of genes that inhibit viral replication. This function is vital for maintaining antiviral state and modulating immune response, making IFN-alpha a key player in both innate and adaptive immunity. Type I interferons, including 14 IFN-α genes, are clustered on human chromosome 9, highlighting evolutionary significance and complexity of interferon signaling pathways. IFN-α, along with IFN-β and IFN-ω, exerts a range of biological effects, including antitumor and immunomodulatory activities, and is increasingly utilized in clinical settings to treat various malignancies, myelodysplasias, and autoimmune diseases. Type I interferon receptor (IFN-αR), which interacts with IFN-α, IFN-β, and IFN-ω, is a multisubunit receptor that underscores intricate regulatory mechanisms governing immune responses.

I read the first review that the researcher use this product for fixed frozen mouse tissues for IF; thus, is it possible use this antibody for western-blot of mouse IFN alpha?

Asked by: jessie011
Thank you for your question. We have not yet tested this antibody in mouse samples, and so it is not recommended for detection in mouse at this time. The exact epitope recognized by this monoclonal antibody has not yet been determined. The full-length human antigen sequence appears to share approximately 65% sequence identity with some of IFN alpha proteins in mouse. It may cross-react, or it may not, but this would have to be determined experimentally.

I am using IFN-α (F-7): sc-373757 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: jenniferc
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
