IL-8 Antibody (C-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects IL-8 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). IL-8 Antibody (C-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Interleukin-8, commonly referred to as IL-8, is a crucial member of the C-X-C chemokine family and serves as a potent chemoattractant cytokine that plays a vital role in the chemotaxis and activation of neutrophils, which are essential for the immune response. IL-8 is primarily secreted by activated macrophages and epithelial cells, and IL-8 expression is significantly associated with various chronic inflammatory diseases, such as inflammatory bowel disease and atherosclerosis. Notably, IL-8 is produced as a 99 amino acid precursor that is cleaved to yield a 72 amino acid, nonglycosylated, biologically active form. The dynamic equilibrium between IL-8 monomers and dimers, both in solution and bound to cell surfaces, is critical for regulating chemotaxis and receptor signaling. Research indicates that IL-8 dimerization acts as a negative regulator of IL-8 receptor function, with two receptors, IL-8RA and IL-8RB, exhibiting 77% sequence identity and functioning as seven-transmembrane domain proteins similar to G protein-coupled receptors. These receptors not only bind IL-8 but also interact with other members of the α and β chemokine families, underscoring IL-8′s importance in mediating inflammatory responses and potential as a therapeutic target in related diseases.

How can IL-8 (C-11): sc-376750 monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody, c-Myc (9E10), is currently available conjugated to phycoerythrin (sc-376750 PE), fluorescein (sc-376750 FITC), Alexa Fluor® 488 (sc-376750 AF488) or Alexa Fluor® 647 (sc-376750 AF647).

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with IL-8 (C-11): sc-376750?

We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems.
