IFN-γRα Antibody (GIR-94) is a mouse monoclonal IgG2b kappa light chain antibody that detects the IFN-gamma R alpha protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). IFN-γRα monoclonal antibody (GIR-94) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The IFN-gamma receptor plays a crucial role in mediating the effects of interferon-gamma, a key cytokine involved in immune responses. Upon binding of interferon-gamma, the receptor undergoes a conformational change that activates associated Janus kinases, leading to the phosphorylation of transcription factors such as Stat1. This process is vital for the transcriptional activation of genes involved in antiviral responses, immune regulation, and cell proliferation. The receptor is composed of two subunits, IFN-γRα and IFN-γRβ, with the latter being essential for signal transduction and the full biological response to interferon-gamma. Notably, while IFN-γRα is sufficient for ligand binding, the presence of both chains is necessary for the activation of IFN-γ-inducible genes, highlighting the importance of their interaction in immune signaling pathways.

Hi! Can sc-12755 be used for IHC staining? Also, is it membrane stained? Thanks in advance!

Thank you for your question. We have not yet tested this antibody, IFN-γRα Antibody (GIR-94): sc-12755, on formalin-fixed paraffin-embedded tissue sections, and so IHC(P) is not a recommended application at this time. However, we do recommend and warranty the antibody for IF, or immunofluorescent staining of fresh or frozen tissue sections or cells fixed in acetone or methanol. We do expect membrane staining for IFN-γRα. If you are looking for an antibody for IHC(P) in human tissues, we would recommend an alternative clone, IFN-γRα Antibody (D-3): sc-28363. Can IFN-γRα (GIR-94): sc-12755 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, IFN-γRα (GIR-94): sc-12755 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Hi, I am interested in using this antibody for Western Blot of mouse cells. However, I checked the same clone (GIR-94) from BD science, they said it is only used for human. Could you let me know if this antibody is suitable in detecting mouse IFNGR1?

Thank you for your question. We have tested this mouse monoclonal, IFN-γRα Antibody (GIR-94): sc-12755, in mouse samples and have verified reactivity with mouse peripheral blood leukocytes in Flow Cytometry. Therefore, we do recommend this antibody for detection of mouse IFNGR1 by Western blotting as well.
