γ-GCSc Antibody (H-5) is a mouse monoclonal IgG2a kappa light chain antibody that detects gamma-GCSc protein, also known as glutamate-cysteine ligase catalytic subunit (GCLC), in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). γ-GCSc monoclonal antibody (H-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The GCLC gene, comprising 16 exons, encodes the 636 amino acid gamma-GCSc protein, which plays a crucial role in glutathione synthesis, a vital antioxidant that protects cells from oxidative stress. Gamma-GCSc is primarily expressed in hemocytes, brain, liver, and kidney, where gamma-GCSc forms a heterodimer with the regulatory subunit gamma-GCSm, collectively known as gamma-GCS. Gamma-GCSc catalyzes the first and rate-limiting step in glutathione biosynthesis, making gamma-GCSc essential for cellular defense against reactive oxygen species and maintaining redox homeostasis. Gamma-GCSc regulation is influenced by various factors, including oxidants and heavy metals, which can enhance transcription, thereby increasing glutathione levels and potentially providing protective effects against chemically induced carcinogenesis. Notably, mutations such as the His370Leu change in gamma-GCSc can lead to significant deficiencies in enzyme activity, resulting in conditions like hemolytic anemia and reduced glutathione levels in red blood cells.
