MLF1 Antibody (A-8) is a mouse monoclonal IgG1 antibody that detects myeloid leukemia factor 1 (MLF1) in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MLF1 is a crucial 268 amino acid protein that plays a significant role in regulating cell cycle progression, particularly as a negative regulator functioning upstream of the tumor suppressor p53. This regulation is vital because p53 is a key player in preventing tumor formation by inducing cell cycle arrest in response to DNA damage. Anti-MLF1 antibody (A-8) promotes p53-dependent cell cycle arrest in murine embryonic fibroblasts and inversely affects the levels of COP1, a ubiquitin ligase that targets p53 for degradation. Furthermore, MLF1′s expression inhibits erythropoietin-induced cell cycle exit and the increase of the cell cycle inhibitor p27, highlighting MLF1′s multifaceted role in cell cycle regulation. Notably, Polo-like kinase 1 (Plk1) phosphorylates MLF1 at the Thr78 site, leading to MLF1′s ubiquitination and degradation during the critical transition from metaphase to anaphase. Mutations at these phosphorylation sites can stabilize MLF1, thereby disrupting normal mitotic progression and potentially contributing to leukemogenesis. Given MLF1′s essential functions in multi-potent progenitor cells, MLF1′s dysregulation is a significant factor in the development of acute myeloid leukemia, making MLF1 (A-8) monoclonal antibody an important tool for research in cancer biology and cell cycle regulation.
