CD32-B Antibody (F-4) is a mouse monoclonal IgG1 antibody that detects CD32-B of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). CD32-B (F-4) antibody is available in a non-conjugated form, providing flexibility for various experimental needs. CD32, also known as Fc gamma RII, serves as a low-affinity receptor for the Fc fragment of aggregated immunoglobulin G (IgG), playing a crucial role in the immune response by mediating the clearance of immune complexes through macrophages. This function is vital for maintaining homeostasis and preventing autoimmune reactions. Additionally, CD32-B is involved in the regulation of antibody production by B cells, where CD32-B interaction with IgG delivers a negative signal that modulates B cell activity. CD32-B exists in several isoforms, produced by alternative splicing of three distinct genes (A, B, and C), which are designated FcgRIIA, FcgRIIB1, FcgRIIB3, and FcgRIIC. These isoforms are expressed on various immune cells, including monocytes, placental trophoblasts, and endothelial cells, with specific forms present on B lymphocytes and neutrophils, highlighting CD32-B′s diverse roles in immune regulation and response.

Rated 4 out of 5 by from Not for use on IHC-p or flow cytometryI requested a sample of this mouse monoclonal F-4 anti-CD32b to test it for unvalidated procedures. I tested whether it would work in flow cytometry by intracellular staining and for immunofluorescent imaging on formalin-fixed paraffin-embedded (FFPE) tissue. Both procedures are greatly dependent on the epitope-specificity of the antibody, and it is not surprising that this clone did not work for either application. What is encouraging is that this antibody showed almost no background (non-specific) binding in either application, suggesting a high degree of specificity. IHC-p procedure: Human spleen section. Deparaffinization, rehydration, heat-mediated epitope retrieval with either citrate pH6 or EDTA pH8, overnight staining with F-4, secondary rat anti-mouse IgG1, then tertiary goat anti-rat IgG-AlexaFluor568. Flow cytometry procedure: PBMCs. Surface stain for IgE, CCR3 and CD3. Fixation and permeabilization using BD reagents. Overnight staining with primary. Secondary rat anti-mouse IgG1-APC. Gating on CCR3+IgE+ (basophils, positive control) or CD3+ (T cells, negative control).
