    GITR Antibody (621) is a mouse monoclonal IgG1 kappa light chain antibody that detects GITR protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), flow cytometry (FCM), and enzyme-linked immunosorbent assay (ELISA). GITR (621) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The glucocorticoid-induced tumor necrosis factor receptor family-related protein (GITR) plays a crucial role in modulating immune responses, particularly in T lymphocytes, where GITR is preferentially expressed in the thymus, spleen, and lymph nodes. GITR is a member of the tumor necrosis factor receptor (TNFR) superfamily, which is characterized by type I transmembrane glycoproteins that are integral to various cellular functions, including proliferation, differentiation, and programmed cell death. The structure of GITR includes a cysteine-rich extracellular domain that facilitates ligand binding and an intracellular death domain that is essential for signal transduction. This structural configuration allows GITR to interact with other proteins in the TNFR superfamily, such as Ox40, 4-1BB, and CD27, thereby influencing the activation of critical signaling pathways like NFκB, which promotes cell survival, and caspase pathways, which lead to apoptosis. Understanding the structure and function of GITR is vital for developing therapeutic strategies aimed at enhancing or inhibiting immune responses in various diseases, including cancer and autoimmune disorders.

    Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, GITR (621): sc-53972 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

    Rated 4 out of 5 by from I would purchase againI received a sample lot D2616 and got great signal in an ELISA, so I ordered another vial. The second lot C1607 didn't work at all. When I called in to tech support I was told that it was an old lot and they would be happy to replace it with a more recent lot I2616. It arrived the next day. It was still not as good as the sample lot that I received, but it did work as my positive control.
