HLA-DMβ Antibody (DM.K8) is a mouse monoclonal IgG1 antibody that detects HLA-DMβ in human samples through applications such as western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). HLA-DMβ, also known as DMB or RING7, is a 263 amino acid single-pass type I membrane protein that plays a pivotal role in the immune response by facilitating the release of class II-associated invariant chain peptide (CLIP) from newly synthesized MHC class II molecules. This critical function allows for the peptide binding site to be available for the acquisition of antigenic peptides, thereby enhancing the presentation of these peptides to T cells. The interaction between HLA-DM and MHC class II molecules is further regulated by HLA-DO, particularly in B cells, highlighting the intricate balance of immune signaling. HLA-DMβ exists as a heterodimer, composed of an alpha chain (DMA) and a beta chain (DMB), which is essential for its function. The gene encoding HLA-DMβ is located on human chromosome 6p21.32 and spans approximately 6,442 bases, underscoring its significance in the major histocompatibility complex (MHC) class II family. With its ability to modulate immune responses, HLA-DMβ is a vital component in understanding antigen presentation and the overall functioning of the adaptive immune system.
