SEE ALSO...

MHC class I Antibody (W6/32) is a mouse monoclonal IgG2a kappa light chain antibody that specifically detects HLA-A, HLA-B, and HLA-C molecules of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). MHC class I monoclonal antibody (W6/32) recognizes a monomorphic epitope present on the heavy chain of human MHC class I molecules, enabling broad detection of these essential cell surface proteins. MHC class I molecules, also known as human leukocyte antigens (HLA) class I, including HLA-A, HLA-B, and HLA-C, are crucial for the immune system′s ability to present endogenously derived peptide antigens to cytotoxic T lymphocytes (CD8+ T cells). By presenting peptides typically 8-10 amino acids in length, MHC class I molecules allow the immune system to monitor cellular health and identify infected or malignant cells. MHC class I molecules function is vital for immune surveillance and initiating cytotoxic immune responses, playing a key role in controlling viral infections and tumor development. Each MHC class I molecule is composed of a heavy alpha chain and a light chain called β2-microglobulin. The heavy alpha chain contains a peptide-binding groove that is essential for antigen presentation to cytotoxic T lymphocytes. The light chain, β2-microglobulin, is non-covalently associated with the alpha chain and is crucial for the proper folding, stability, and surface expression of the MHC class I complex. Dysregulation or downregulation of MHC class I expression can lead to immune evasion by pathogens or cancer cells, highlighting their importance in disease processes. MHC class I (W6/32) antibody serves as a valuable tool for researchers studying antigen presentation, immune recognition, and related immunological functions.

How much AF 647 antibody do you use for Flow Cytometry?

Asked by: 12345678987654321
Thank you for your question. We recommend using 1 µg of fluorochrome-conjugated antibodies to 100 µl of the prepared cell suspension (equal to 1 million cells). https://www.scbt.com/resources/protocols/flow-cytometry

Are the agarose conjugated antibodies covalently bound ? Do they resist to acidic elution for instance and keep attached to the agarose? Thx

Asked by: YannE
Thank you for your question. Yes, they are covalently bound via the primary amine. I do not have information specifically regarding acidic elution.

I want an antigen which can not react with β2 microglobulin existing in the β chian. Does it react with all regions of MHC class I, or just react with α region?

Asked by: Sarah Wang
Thank you for your question. This antibody recognizes the W6/32 antigenic determinant common to HLA-A, B and C antigens of human origin. The exact epitope for this antibody has not been epitope mapped, so we do not have a more detailed answer. If you have any further questions, please contact our Asia Technical Service team.
