caspase-8 Antibody (8CSP03) is a mouse monoclonal IgG1 kappa light chain antibody that detects the caspase-8 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). anti-caspase-8 antibody (8CSP03) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Caspase-8 plays a crucial role in the apoptotic signaling pathway, acting as an initiator caspase that activates downstream effector caspases through the cleavage of their inactive precursors. This process is vital for the regulation of programmed cell death, particularly in response to death receptor signaling via tumor necrosis factor receptor superfamily member 1A (TNFRSF1A) and Fas (TNFRSF6). Caspase-8 is primarily located in the cytoplasm and is recruited to the death-inducing signaling complex (DISC) upon binding to the adaptor molecule FADD, where activation occurs. The active form of caspase-8, a heterodimer composed of p18 and p10 subunits, is then released from the DISC to propagate the apoptotic signal. Caspase-8 is widely expressed, with the highest levels found in peripheral blood leukocytes, thymus, liver, and spleen. Mutations or defects in the CASP8 gene can lead to caspase-8 deficiency disorder (CASP8D), which is associated with splenomegaly and impaired apoptosis of peripheral blood leukocytes, potentially resulting in immunodeficiency due to compromised activation of T lymphocytes, natural killer cells, and B lymphocytes.

Thank you for your question. Most of our antibodies are only tested in samples from mouse, rat, and human. We unfortunately do not have information on reactivity of our caspase 8 antibodies with the drosophila homolog.

Can I use this mouse monoclonal antibody for immunohistochemical staining of mouse tissue? Or even double or triple staining on the same samples?

Rated 4 out of 5 by from Acceptable for detection non-cleaved Caspase8 (WB)Antibody specifically recognizes non-cleaved Caspase 8 , however detection in lysates of HCT116 cells was rather week. I recommend to use fresh lysates and load more ug of protein on gels.
